This issue is fixed in WebLink 9 SP1. Please see KB 1013665.
Does this error only occur when you are testing the SMTP settings? If you just click OK and save the settings regardless, then try to email a document in WebLink, do you get a similar error?
Your mail server might require authentication. If you check "SMTP requires authentication", does it work?

Use the same information on another mail client and see if it works independent of Weblink. Make sure the account you are using to access the mail server actually has rights to send email from this mail server.

We're looking into a possible issue where testing the SMTP configuration in the WebLink Administrator's Utility always uses WebLinkAutomatedSender@Laserfiche.com instead of what you've actually configured as the reply address.
For now though, if you just save your SMTP configuration and then log into WebLink and send a document as an email, is that successful?
