Have you created a starting rule on that folder? In the Rule Manager tab in Workflow Designer, select the workflow you want and it's starting rules should show up on the right side. You'll then have the option to add or make changes.

If you already have the starting rule created, you can check to see how they are being evaluated by opening the Workflow Administration Console and looking at the Subscriber Trace Log.

Devin is spot on--you'll want to set up your starting rules such that Workflow is monitoring your Auto-File folder. The Workflow help files has a section specifically on starting rules. It goes into detail on creating and editing them. I highly suggest taking a look at it.

You can also start your Workflow from the designer if you're using Workflow 9 or later. Again, the help files do an excellent job of explaining how to use this tool.
